Determination of cardinal temperatures for sugar apple ( Annona squamosa L.) Ciência e Agrotecnologia
Rodrigues,Bruno Rafael Alves; Santos,Rayane Carneiro dos; Nietsche,Silvia; Mercadante-Simões,Maria Olívia; Cunha,Isabella Renata Gomes da; Pereira,Marlon Cristian Toledo.
ABSTRACT The sugar apple is highly dependent on the pollination and fruit tree pollen performance is crucial for successful fertilization and fruit set. The objective of this study was to determine the cardinal temperatures for in vitro pollen grains germination and evaluate the effect of temperature on pollen tube growth of sugar apple. The experimental design was in a completely randomized with eight treatments (10, 15, 20, 25, 30, 35, 40 and 45 ºC), four replicates and each plot was constituted by two slides per parcel. The cardinal temperatures were determined by evaluating in vitro germination of pollen grains and pollen tube growth in standard culture medium. It also assessed the pollen tube growth and the percentage of germination in vitro depending...

Floral phase and sucrose/boric acid concentrations on in vitro germination of Inga edulis (Mart.), pollen grain Sci. Agrar.Paran. / SAP
Santos, Ana Claudia Schllemer dos; Castro, Juliana Dias de; Bressan, Daiane; Júnior, Américo Wagner; Guollo, Karina.
The aim of this work was to determine the ideal stage of pollen collection and the best concentrations of sucrose and boric acid in the culture medium for the in vitro germination of inga fruit tree pollen. For this, the work was carried out in two stages, one using concentrations of sucrose in the culture medium (0, 5, 10, 20 and 30%) - experiment 1. Another one using the best sucrose concentration, with concentrations (0, 100, 200 and 300 mg L-1) - experiment 2. In the two experiments, two flowering phases were use: pre-anthesis and post-anthesis. For both experiments, the culture medium contained 1% agar to solidify the medium and all treatments remained in BOD for 24 h at a temperature controlled at 25°C. For the experiment 1 with sucrose, DIC was use...

Histological analysis of pollen-pistil interactions in sour passion fruit plants (Passiflora edulis Sims) Biocell
Madureira,Hérika Chagas; Pereira,Telma Nair Santana; Cunha,Maura Da; Klein,Denise Espellet.
The success of sexual plant reproduction is directly influenced by specific interactions between the pollen and pistil. Light, fluorescence and scanning electron microscopy techniques were used to evaluate the steps of pollination in sour passion fruit plants (Passiflora edulis Sims). In the compatible interaction, pollen tubes grow through stigma projections towards the ovary. The pollen grain surface was found to be spheroidal and to consist of heteroreticulate exine with six colpi. Furthermore, analysis in vivo of pollen-pistil interactions indicated that stigmas of flowers 24 hours before anthesis are unable to discriminate compatible (genetically unrelated) and incompatible (genetically related) pollen grains. Taken together, these results provide...

Morphology and viability of pollen grains from passion fruit species (Passiflora spp.) Acta Botanica
Soares,Taliane Leila; Jesus,Onildo Nunes de; Souza,Everton Hilo de; Santos-Serejo,Janay Almeida dos; Oliveira,Eder Jorge de.
The characterization and viability of pollen grains are useful tools to guide crosses in breeding programs. The objective of this study was to describe the morphological patterns and viability of pollen grains from five accessions of Passiflora edulis f. flavicarpa O. Deg. and five accessions of Passiflora setacea DC. Pollen morphology descriptions were made using light microscopy and scanning electron microscopy, whereas the viability analysis was performed by in vitro germination and histochemical analysis (Lugol's solution and 2,3,5-triphenyltetrazolium chloride). Pollen grains assessed for germination were inoculated in culture medium containing Ca(NO3).4H2O (0.03%), Mg(SO4).7H2O (0.02%), KNO3 (0.01%), H3BO3 (0.01%), sucrose (15%) and agar (0.8%)....

Structural cell wall proteins from five pollen species and their relationship with boron Braz. J. Plant Physiol.
García-Hernández,Edith del R.; López,Gladys I. Cassab.
Boron (B) is an essential micronutrient for the survival of vascular plants. The most severe effect derived from a deficiency of B is the alteration of cell wall biogenesis and pollen germination. We investigated pollen of plant species that require B for germination (Zea may L. and Nicotiana tabacum L.), as well as those that can germinate without B (Pinus sp, Lilum longiflorum, Impatiens sp.). In both groups, B addition in the growth medium increased the length of the pollen tube after germination. Hydroxyproline Rich Glycoproteins (HRGPs) are the most abundant cell wall structural proteins of dicotyledonous plants and the sexual tissues of monocotyledonous plants. Here, we show that maize pollen accumulated a significant pool of hydroxyproline (Hyp) and...

Ultrastructural features of Mimulus aurantiacus (Scrophulariaceae) pollen tubes in vivo Anais da ABC (AABC)
Ekici,Nuran; Dane,Feruzan; Olgun,Göksel.
The aim of this study is to give information on ultrastructure of in vivo pollen tubes of Mimulus aurantiacus which were collected from the Botanical Garden of the University of California at Berkeley. Materials were prepared according to electron microscopy methods and examined under Zeiss electron microscope. Four zones were examined in the pollen tubes of Mimulus aurantiacus. APICAL ZONE: Mitochondria, smooth endoplasmic reticulum, rough endoplasmic reticulum, dictyosomes and secretory vesicles were observed. SUBAPICAL ZONE: This area contained abundant rough endoplasmic reticulum and occasionally some smooth endoplasmic reticulum. The polysomes, mitochondria, proplastids that contain starch, small vacuoles and a few lipid bodies were detected. NUCLEAR...
